研究内容

薬学の重要な役割の中に,病気の治療,予防を目的とする医薬品の創薬があります。では,どのようにして医薬品は創り出されるのでしょうか。ほとんどの場合,その基本となるリード化合物が,まず植物や動物から抽出・分離されて,その化学構造上の知見や生物活性に関する情報が提供されます。その結果,より汎用性のある医薬品が合成され,そして,世に送り出されます。

当研究室では,医薬品の素材として,植物やキノコに含有する新しい成分の化学構造を明らかにすると共に,生理活性を有する天然有機化合物を検索することを志向した以下の研究を行っています。

1.生理活性を有するイリドイド成分の検索
イリドイド配糖体およびセコイリドイド配糖体は日本薬局方に収載されているセンブリ,リュウタン,サンシシ,サンシュユなどの主成分であり,生理活性を有する化学成分として知られています.また,生理活性アルカロイドの構成成分でもある有用な天然有機化合物です。当教室ではこれまでに,モクセイ科のイボタノキ属,モクセイ属,ガマズミ属の植物,さらに,センブリ,リュウタン,ゲンチアナ,サンシシなどから数多くの新規イリドイド配糖体およびセコイリドイド配糖体を単離し,それらの化学構造を明らかにしました。また,近年,センブリなどから得られたセコイリドイド配糖体に神経突起伸展活性を有することを見いだしました。

2.薬用に用いられるキク科植物の成分分析
キク科植物にはテルペノイド成分が多く含まれており,古くから香料や薬用とされているものも少なくありません。しかし,これらの研究は類似構造や立体異性体の分離が困難であることから停滞していました。当教室では,近年めざましい発展を遂げている最新のクロマトグラフィーをキク科植物のテルペノイド成分の分析に適用し,これまで不可能であった混合物や立体異性体の分離に成功し,成果をあげています。

3.キノコに含まれる生体機能調節物質の検索
キノコは古くから薬用や食用として広く用いられており,医食同源の観点から近年見直されています。当教室では種々の新規ステロールが薬用キノコ,食用キノコおよび毒キノコに含有されていることを見いだしました。これまでに約30種のキノコについて成分分析を行い,数多くの新規ステロール類の分子構造を明らかにし,さらに,それらの機能性について検討しています。
