研究内容
 
当教室では中枢薬理学の観点から、以下のテーマについて研究を進めている。
1. 精神神経疾患モデル動物の作製とその発症機序の解明
現在までに以下に示す3種の精神神経疾患モデル動物の作製を確立し、その発症機序を行動薬理学的、生化学的ならびに病理組織学的手法を用い明らかにしている。
@ アルツハイマー型認知症モデル;マウスを幼若期からビタミンB1欠乏食あるいは低タンパク食で長期間飼育すること、およびマウスの嗅球摘出により記憶・学習障害が観察されさらにこれらの動物では記憶・学習障害といった中核症状のみならず、うつ等の周辺症状も伴っており、アルツハイマー型認知症モデルとして非常に妥当性を有していることを明らかにしている。
A 統合失調症モデル;幼若期ラットの腹側海馬にイボテン酸処理することにより破壊すると思春期後に認知障害が誘発され、この動物は統合失調症モデルとして広く受け入れられている。しかしながら、この明確な発症機序は解明されていないので、この点について詳細に検討を行っている。
B 注意欠陥/多動性障害モデル;断続的にレム断眠をマウスに施すことにより自発運動量の亢進や高頻度の跳躍が誘発し、これらの行動は注意欠陥/多動性障害の治療に用いられるメチルフェニデートによって抑制されることから断続的レム断眠マウスの注意欠陥/多動性障害モデルとしての妥当性を示唆している。

2. 脊髄疼痛伝達・制御機構におけるダイノルフィン系の役割の解明
ダイノルフィンをマウスの脊髄クモ膜下腔内へ投与した際、高用量では抗侵害刺激作用を示すのに対し、極めて低用量では侵害刺激行動を誘発することを見出した。これらのダイノルフィンの作用について検討を行った結果、抗侵害刺激作用の終結にはシステインプロテアーゼとエンドペプチダーゼ24.11が関与すること、侵害刺激行動の発現はNMDA受容体イオンチャネル複合体のポリアミン調節部位の活性化に起因することを明らかにしている。
